In mammals Musashi is important for spermatogenesis and male fertility, but its role in the ovary has yet to be characterized. In this study we determined the expression of mammalian Musashi proteins MSI1 and MSI2 during mouse folliculogenesis, and through the use of a MSI2-specific knockout mouse model we identified that MSI2 is essential for normal follicle development. Time-course characterization of MSI1 and MSI2 revealed distinct differences in steady-state mRNA levels and protein expression/localization at important developmental time-points during folliculogenesis. Using a gene-trap mouse model that inactivates Msi2, we observed a significant decrease in ovarian mass, and change in follicle-stage composition due to developmental blocking of antral stage follicles and pre-antral follicle loss through atresia. We also confirmed that hormonally stimulated Msi2-deficient mice produce significantly fewer MII oocytes (60.9% less than controls, p < 0.05). Furthermore, the majority of these oocytes are of poor viability (62.2% non-viable/apoptotic, p < 0.05), which causes a reduction in female fertility evidenced by decreased litter size in Msi2-deficient animals (33.1% reduction to controls, p < 0.05). Our findings indicate that MSI1 and MSI2 display distinct expression profiles during mammalian folliculogenesis and that MSI2 is required for pre-antral follicle development.]]> Wed 11 Apr 2018 16:39:10 AEST ]]> Oolemmal proteomics : identification of oocyte cell surface protein complexes involved in murine fertilisation https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:2940 Wed 11 Apr 2018 16:05:16 AEST ]]> Mouse Emi2 is required to enter meiosis II by reestablishing cyclin B1 during interkinesis https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:6545 Wed 11 Apr 2018 14:52:54 AEST ]]> Investigation of equine fertility mechanisms through proteomic analysis of stallion spermatozoa, early equine embryos and pregnant mare uterine fluid https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:27549 Wed 11 Apr 2018 14:20:32 AEST ]]> Characterisation of an oocyte specific knockout model of Cdh1 https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:11553 Wed 11 Apr 2018 14:19:59 AEST ]]> Impact of periconceptional and preimplantation undernutrition on factors regulating myogenesis and protein synthesis in muscle of singleton and twin fetal sheep https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:27351 Wed 11 Apr 2018 13:32:44 AEST ]]> Susceptibility of mammalian oocytes to chromosome segregation errors with maternal aging https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:21949 Wed 11 Apr 2018 13:00:11 AEST ]]> The molecular basis of sperm - oocyte interactions https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:10297 Wed 11 Apr 2018 12:39:27 AEST ]]> Premature dyad separation in meiosis II is the major segregation error with maternal age in mouse oocytes https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:21342 Wed 11 Apr 2018 12:17:52 AEST ]]> The impact of maternal ageing and chemotoxicants on female fertility https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:27104 Wed 11 Apr 2018 11:47:19 AEST ]]> Oxidative stress and male reproductive health https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:21426 Wed 11 Apr 2018 11:46:49 AEST ]]> Advances in assisted reproductive techniques for the conservation of Australian carnivorous marsupials https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:5839 Wed 11 Apr 2018 10:16:08 AEST ]]> Fetal programming in 2-year-old calving heifers: peri-conception and first trimester protein restriction alters fetal growth in a gender-specific manner https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:17477 0.1).
